超甜玉米胚性愈伤组织诱导体系的建立 被引量:8

摘要 以超甜玉米自交系S1和品种粤甜3号为材料研究了影响愈伤组织诱导和胚性形成的几个关键因素。结果表明,2种基因型都是高诱导力的材料,自交系S1能够诱导出高质量的Ⅱ型胚性愈伤,是理想的基因转化材料。2,4-D浓度为2 m g/L时,2种基因型出愈率和胚性愈伤率达到最高,胚性愈伤率分别达到45.88%和35.00%;在培养基中添加9 m g/L的A gNO3极显著地提高了胚性愈伤率,比对照提高了31.25%;添加700 m g/L的脯氨酸,胚性愈伤率显著地提高了12.41%。据此建立了高效稳定的超甜玉米转基因胚性愈伤组织诱导体系。 Effect of a few key factors on Supersweet corn immature embryo callus induction and somatic-embryoid formation were studied. The results showed that, both genotypes were high induction materials, and the inbred line S1 was an ideal transgenic material for the highest inducing frequency of the type Ⅱ embryo callus. When the most suitable concentration of 2,4-D was 2 mg · L^-1, the highest rates of embryoid-callus formation of the inbred line, S1, and a cultivar, Yuetian 3, were 45.88% and 35.00%, respectively. The addition of 9 mg · L^-1 of AgNO3 to the N6 medium significantly stimulated embryoid formation and the rate of embryogeny was increased 31. 25% compared to that of control. Furthermore, the addition of L-proline (700 mg · L^-1), which regulated osmotic pressure and promotes the process of embryogeny, enabled the frequency of embryoid formation to be increased 12.41% compared to that of control. A stable, high efficient embryogenic callus induction system has been established in supersweet corn.
