摘要
目的研究去铁胺对K562细胞的抗增殖活性及细胞周期的作用。方法用K562细胞作为人类红白血病细胞株,用MTT法观察DFO的抗增殖作用;流式细胞术分析细胞周期的变化。结果小剂量DFO(12.5μM)处理K562细胞时,生长曲线轻微变化,但是中剂量和大剂量DFO(25μM,50μM)干预后,生长曲线高峰显著低于对照组。DFO抗细胞增殖活性为剂量依赖性。用DFO(25μM,50μM)处理K562细胞48h和72h后,G0/G1期细胞数量增加,S期细胞数量减少,与对照组比较均有显著性差异(P<0.001)。结论去铁胺通过阻止细胞由G0/G1期进入S期发挥抗K562细胞的增殖作用。
Objective To investigate the effect of Desferrioxaminc on antiproliferative activity and the cell cycle of cultured K562 cell. Methods Using K562 cell as human leukemia cell lines, the antiproliferative effect of desferrioxamine (DFO) was observed by MTT, and the cell cycle analysis was performed by flow cytometry. Results When K562 cells were treated with low - dose DFO( 12.5 μM), the growth curve changed slightly, but the peaks of the growth curve were lower significantly than that of the control group when K562 cells were treated with mediate - dose ( 25 μM) and highdose DFO(50μM). The antiproliferative activity of DFO increased in a dose dependent manner. The proportion of cells in G0/G1 phase increased and that in S phase decreased, and was significantly different from that of the control group( P 〈0. 001 ) when K562 cells were treated with DFO (25μM, 50μM) after 48 hours and 72 hours. Conclusion The antiproliferative activity of desferrioxamine was caused by blocking the entrance of cells from G0/G1 to S phase of the cell cycle.
