摘要
目的建立一次实验就能检测出同一样品中的多个病原细菌的基因芯片。方法检索相关细菌的16 s rRNA序列,使用生物信息学软件针对每个细菌的四个可变区设计出4条寡核苷酸探针并制成基因芯片;利用此芯片对6种临床常见病原细菌进行检测。结果6种临床常见病原细菌的检测结果中除铜绿假单胞菌两条探针较弱外,其余5种均能正确与自己的探针杂交,并且混合样品检测结果达到了预期的目标。结论利用寡核苷酸芯片系统实现病原菌通用检测是完全可行的,对部分实验菌株进行初步考核,具有理想的特异性和灵敏度,可以进一步用于临床标本的检测。
Objective To develope a gene chip for determination ot multl-analytes m one sample at the same time, Methods Hundreds of bacteria 16S rRNA.sequences were retrieved and analyzed, four specific oligonucleotide probes of each becteria were designed by using of ,some bioinformatics software. These probes were employed to make gene chips by spoter. Six kinds of clinical infectious bacteria were tested by using this gene chip. Results Among the results of detection of 6 clinical infectious bacteria,5 clinical infections bacteria could he connected exactly with own probe and most of the bacteria could be determined by four specific oligonucleotide probes. Conclusion The result of this research shows that the system of oligonucleotide chips can be used on the universal detection of pathogenic bacteria. Some bacteria for experiment have been used to show that the chip is perfectly specific and sensitive ,so the further research for clinical sample is feasible.
