摘要
目的研究人野生型p53基因对人结肠癌SW1116细胞生长的影响。方法采用电穿孔法将正向携带有野生型p53基因cDNA的重组反转录病毒质粒导入有p53基因突变的人结肠腺癌SW1116细胞,筛选带外源p53基因的G418抗药性SW1116细胞克隆,对其进行生长速度、软琼脂集落形成及裸鼠成瘤等方面的检测,分析其生物学特性变化。结果导入人野生型p53基因使人结肠腺癌SW1116细胞的生长速度明显减慢(P<0.05或0.01)、软琼脂集落数量明显较对照组减少(P<0.05或0.01),移植瘤体积较对照组明显小(P<0.05)。结论人野生型p53基因对人结肠腺癌SW1116细胞有明确的抑瘤效应。
Objective To evaluate the effects of wild-type p53 gene on the growth of human colorectal carcinoma SW1116 cells. Methods Recombinant retroviral vector (Dol-p53) carrying wild-type p53 gene was transfered into SW1116 cells, the cell growth curve, soft agar colony formation and tumorigenesis in nude mice of transfected SW1116 cells were evaluated and the change of their biologic characteristics was also analyzed. Results The transfection of wild-type p53 gene into SW1116 ceils can suppress the phenotype of SW1116 cells, including inhibition of proliferation in culture, anchorage-independent growth and tumorigenecity. Conclusion Wild-type p53 gene can inhibit the growth of human colorectal carcinoma ceils.
