摘要
目的:观察以荔枝为主要原料的复方口服液舒糖宝对糖尿病大鼠血糖、血脂、尿糖、尿蛋白的干预效应。方法:①实验于2004-10-05/10-22在广西右江民族医学院生化教研室实验室完成。选用健康雄性Wistar大鼠18只。随机将大鼠分为3组:模型组、治疗组、对照组,每组6只。②模型组和治疗组剂量按180mg/kg剂量腹腔注射四氧嘧啶,一次注完。注射后第3,7天各测一次空腹血糖。血糖升高稳定后,并达15mmol/L以上为造模成功。造模成功后治疗组开始用舒糖宝口服液(主要成分为本地酸荔枝、沙田柚、番石榴等)按0.5mL/只灌胃,1次/d,连续10d。模型组和对照组(未造模)用等体积生理盐水灌胃,1次/d,连续10d。③血糖用邻甲苯胺微量法测定,三酰甘油采用GPO-POD酶法测定,总胆固醇采用COD-CE-PAP酶法制定,高密度脂蛋白胆固醇水平测定采用沉淀法测定,尿蛋白用双缩脲法测定,尿糖用葡萄糖氧化酶法测定。④计量资料差异比较采用方差分析。结果:大鼠18只均进入结果分析。①空腹血糖:造模前模型组大鼠明显高于对照组(P<0.01),治疗组明显低于对照组(P<0.01);造模3d后,模型组和治疗组明显高于对照组(P<0.01);舒糖宝治疗3和10d后,模型组和治疗组明显高于对照组(P<0.01),模型组明显高于治疗组(P<0.05,0.01)。②血清三酰甘油水平:舒糖宝治疗10d后,对照组和治疗组明显低于模型组(P<0.01),对照组明显低于治疗组(P<0.05)。③血清总胆固醇水平:模型组和治疗组明显高于对照组(P<0.01),模型组明显高于治疗组(P<0.05)。④高密度脂蛋白胆固醇水平:治疗组明显高于模型组和对照组(P<0.01)。⑤尿蛋白:治疗组舒糖宝治疗7和10d后和对照组明显低于模型组(P<0.01),模型组舒糖宝治疗10d后明显低于治疗7d后(P<0.01),治疗组舒糖宝治疗7d后明显高于治疗前(P<0.01),治疗组明显低于模型组,明显高于对照组(P<0.01)。⑥尿糖水平:模型组和治疗组治疗前及舒糖宝治疗7,10d后明显高于对照组(P<0.01),模型组治疗前明显低于治疗组(P<0.01),治疗10d后明显高于治疗7d后(P<0.01),治疗组治疗7,10d后明显低于治疗前(P<0.01),且治疗10d后低于治疗7d后。结论:①舒糖宝具有降低四氧嘧啶诱导的糖尿病大鼠血糖水平,调查血脂紊乱状况功能。②舒糖宝对糖尿病大鼠肾脏功能有一定的保护作用。
AIM: To observe the intervention effect of shutangbao on blood glucose, blood lipid, glucose and urine protein in rats with diabetes.
METHODS: (1)The experiment was conducted in the Laboratory of Stuff Room of Biochemistry, Youjiang Medical College for Nationalities froria October 5^th to October 22^th 2004. 18 healthy male Wister rats were randomly divided into 3 groups: Model group, treatment group and control group, with 6 rats in each group. (2)Rats in the model group and treatment group received intraperitoneal injection of alloxan at a dose of 180 mg/kg in one time. Fasting blood glucose were measured at the 3^rd and 7^th days of injection. Modeling was made successfully when the blood glucose stablely increased to 15 mmol/L. Rats in the treatment group orally took shutangbao (main components: Acid litchi, grapefruit and guava etc.) after modeling with gastric perfusion of 0.5 mL in each rat, once a day for 10 continuous days. Rats in the model group and normal control group received gastric perfusion of normal saline of the same volume, once a day for 10 continuous days. (3) Level of blood glucose was measured with toluidine tracing method, level of triacylglycerol was measured with GPO-POD enzyme method, level of total cholesterol was assayed with COD-CE-PAP enzyme method, level of high density lipeprotein (HDL) cholesterol was assayed with sedimentation, level of urine pertein was assayed with biuret method, and level of glucose was assayed with microcide method. (4) Comparison of differences in measurement data was analyzed with analysis of variance.
RESULTS: A total of 18 rats were involved in the analysis of results.(1) Fasting blood glucose: It was significantly higher in the model group before modeling than that in the control group(P 〈 0.01), and it was significantly lower in the treatment group than that in the control group (P 〈 0.01); Three days after modeling, it was higher in the model group and treatment group than that in the control group (P 〈 0.01); At the 3^th and 10^th day of taking shutangbao, it was significantly higher in the model group and treatment group than that in the control group (P 〈 0.01), and it was significantly higher in the model group than that in the treatment group(P 〈 0.05,0.01). (2)Level of serum triacylglycerol: 10 days after taking shutangbao, it was significantly lower in the control group and treatment group than that in the model group (P 〈 0.01 ), and it was significantly lower in the control group than that in the treatment group (P 〈 0.05).(3) Level of serum total cholesterol: It was significantly higher inthe model group and treatment group than that in the control group(P 〈 0.01), and it was significantly higher in the model group than that in the treatment group(P〈0.05). (4) Level of HDL cholesterol: It was significantly higher in the treatment group than that in the model group and control group (P 〈 0.01 ). (5) Level of urine protein: At the 7^th and 10^th day of taking shutangbao, it was significantly lower in the control group than that in the model group(P 〈 0,01 ), and it was significantly lower in the model group at the 10^th day that than that at the 7^th day(P〈0.01 ) , and significantly higher at the 7^th day than before the treatment (P〈0.01 ), and it was significantly lower in the treatment group than that in the modle group and higher than that in the control group (P 〈 0.01 ). (6) Level of glucose: It was obviously higher in the model group and treatment group before the treatment and at the 7^th Sand 10^th day of taking shutangbao than that in the control group (P 〈 0.01 ), and it was obviously lower in the model group before the treatment than that in the treatment group (P 〈 0.01 ), and hi.gher at the 10^th day than that at the 7^th day (P 〈 0.01), and it was significantly lower at the 7^th and 10^th day than that before treatment (P 〈 0.01 ), and lower that at the 10^th day than that at the 7^th day.
CONCLUSION: (1) Shutangbao can reduce the level of blood glucose in rats with diabetes induced by alloxan as well as investigate disorder of blood lipid. (2)Shutangbao has some protective effect on the function of kidney in rats with diabetes.
