巴戟天对过度训练大鼠心肌细胞凋亡的影响 被引量:3

摘要 目的:观察巴戟天对过度训练大鼠心肌细胞凋亡的影响。方法:实验于2003-09/2004-07在郑州大学医学院药理教研室完成。选择SD大鼠45只,大鼠随机分为安静对照组,过度训练组,过度训练+巴戟天组,每组各15只。过度训练组、过度训练+巴戟天组大鼠均采用递增负荷游泳训练建立过度训练模型,每周训练6d,共7周,过度训练+巴戟天组大鼠同时于训练前1h灌服巴戟天4.5mg/(kg·d)。安静对照组常规饲养7周,不加干预。7周后分别取3组大鼠心脏,剪碎后消化离心,制成细胞涂片,采用原位末端标记技术检测大鼠心肌细胞的凋亡率。以匀浆起将心脏制成10%心肌组织生理盐水匀浆,测定超氧化物歧化酶、丙二醛含量。结果:各组大鼠均进入结果分析,无脱失值。①巴戟天对心肌细胞凋亡率的影响:原位末端标记技术结果显示安静对照组心肌细胞偶见阳性核染色标记。过度训练组心肌细胞有较多的阳性标记。过度训练+巴戟天组只有散在的阳性标记,其阳性细胞凋亡率明显低于过度训练组[(6.80±1.99)%,(11.40±2.32)%,(P<001)]。②巴戟天对心肌细胞超氧化物歧化酶和丙二醛含量的影响:过度训练组大鼠心肌细胞超氧化物歧化酶活性显著低于安静对照组和过度训练+巴戟天组[(2.61±0.21)mkat/g,(5.10±0.21,3.89±0.15)mkat/g,(P<0.01)],丙二醛含量则显著高于安静对照组和过度训练+巴戟天组[(3.67±0.17)μmol/g,(1.54±0.11,2.09±0.16)μmol/g,(P<0.01)],过度训练+巴戟天组与安静对照组比较无显著差异。结论:过度训练导致大鼠心肌细胞凋亡显著增加,巴戟天通过抗氧化作用抑制心肌细胞过度凋亡,发挥心肌保护作用。 AIM: To observe the effects of medicinal indianmulberry root on apoptosis of cardiac myocytes in overtrained rats. METHODS: The experiment was conducted at the Department of Pharmacology, Medical College of Zhengzhou University from September 2003 to July 2004. Forty-five SD rats were selected and randomly divided into control group, overtraining group, and overtraining and medicinal indianmulberry root group with 15 rats in each group. The rats in the overtraining group and overtraining and medicinal indianmulberry root group were all treated with progressively the load swimming training to build training models, six days each week for 7 weeks. The rats in the overtraining and medicinal indianmulberry root group' were perfused with 4,5 mg/kg per day medicinal indianmulberry root one hour before training. Those in the control group were fed for 7 weeks routinely without intervention. Rat hearts in the 3 groups were gained after 7 weeks, and then cut into piece and digested with centrifuge to make cell smear. The apoptosis rate of the rat cardiac muscle cells was detected by in situ endlabeling (ISEL) technology. The cardiac muscle was made into saline liquid for 10% by homogenization. The contents of superoxide dismutase (SOD) and malondialdehyde (MDA) were tested. RESULTS: All the rats in every group were involved in the result analysis, without drop-out rats. (1) Effects of medicinal indianmulberry root on apoptosis rate of myocardial cells: The ISEL showed the positive nucleus appeared in myocardial cells of control group by chance. Many positive marks appeared in myocardial cells of overtraining group. The positive labeling appeared diffusedly in the overtraining and medicinal indianmulberry root group, and the positive apoptosis rate was lower significantly than that in the overtraining group [(6.80±1.99) %, (11.40±2.32) %, (P〈0 01)]. (2) Effects of medicinal indianmulberry root on contents of SOD and MDA of myocardial cells: The activity of SOD of myocardial cells in the overtraining group was lower obviously than that in the control group and the overtraining and medicinal indianmulberry root group [(2.61±0.21) mkat/g,(5.10±0.21,3.89±0.15) mkat/g, (P〈0.01)]. The content of MDA was higher markedly than that in the control group and the overtraining and medicinal indianmulberry root group [(3.67±0.17) μmol/g, 1.54±0.11,2.09±0.16)μmol/g, (P〈0.01)]. There was no distinctly difference between" the overtraining and medicinal indianmulberry root group and the control group. CONCLUSION: The overtraining lead to the apoptosis ratio increasing significantly in myocardial cells of rats. The medicinal indianmulberry root can inhibit the ovcr-apoptnsis of myocardial cells by its anti-oxidation and protect myocardial cells.
