空斑法筛选具有中和活性的抗衣原体粘连蛋白McAb

Selection of Neutralizing McAb 18 kDa Binding Protein of Chlamydia Trachomatis Serovar LGV2 Using Plaque Test

本研究制备了抗１８ｋＤａ粘连蛋白的单克隆抗体，并证实在不同血清型之间，甚至不同种衣原体之间１８ｋＤａ粘连蛋白具有一定的共同抗原表位。我们建立的空斑形成试验和单克隆抗体空斑减少中和试验，实验结果明显、客观，易于判定，稳定性好。用此筛选出县有中和活性的２Ｂ９ＭｃＡｂ。该ＭｃＡｂ不同于目前已报道的ＭｃＡｂ，其中和活性不依赖于补体，而可能是抑制衣原体的粘附。

In this study we have produced three McAbs against binding protien of chlamydia trachomatis. All the three McAbs could not only react with LGV2 serovar, but also with LGV1,D and I serovars and TWAR strain,suggesting that 18 kDa proteins of different serovars have common epitops. Using plaque technique we have established that LGV 2 cellular infection model which makes the evaluation of the LGV2 infection,is objective, stable and simple. A plaque inhibition test was performd using McAbs, and the results showed that there was neutralization activity with 2B9, but not with 1A8 and 1D9. Furthermore,unlike McAbs described by others,2B9 showed neutralization activity when complements was absent,suggesting that its activity was probably due to inhibitin of chlamydia adherance to host cells by blocking sites,but not due to complements mediated direct lysis of chlamydia. This study might provided us a new approach to the prevention and treatment of chlamydial diseases.
