芪棱汤对脑缺血-再灌注大鼠Caspase-3蛋白表达的影响

The Effects of Qileng Decoction(芪棱汤) on the Expression of Gasepase-3 Protein in Brain of Rats after Cerebral Ischemia-Reperfusion

目的观察芪棱汤对脑缺血再灌注大鼠Caspase-3蛋白表达的影响。方法采用颈内动脉线栓加环扎法复制大鼠脑缺血再灌注模型,于再灌注后不同时间点采用免疫组化法检测大鼠Caspase-3蛋白表达,采用TUNEL法检测凋亡细胞数。结果补阳还五汤组与芪棱汤组Caspase-3蛋白表达明显低于模型组,芪棱汤组Caspase-3蛋白表达明显低于补阳还五汤组;补阳还五汤组及芪棱汤组均能减少缺血半暗带神经元细胞的凋亡,而以芪棱汤组尤为显著。结论芪棱汤可以通过抑制Caspase-3蛋白表达,减少神经细胞的凋亡,达到神经保护作用。

Objective:To study the effect of Qileng Decoction(芪棱汤) on the expression of Casepase -3 protein in on brain of rats after cerebral ischemia reperfusion. Methods:Middle cerebral aretery occlusion reperfusion(MCAO/IR) model in rat was made by thread emboliging and fastening method. Caspase-3 protein were observed by immunohistochemical method,TUNEL was employed to measure the cell apoptosis at different time point after ischemia reperfusion. Result:The expression of Caspase-3 protein in model group was significantly higher than that in both Buyanghuanwu Decoction(补阳还五汤) group and Qileng Decoction group,and the expression of Caspase-3 in QiLeng decoction group were reduced significantly compared with Buyanghuanwu Decoction group. Both Buyanghuanwu Decoction and Qileng Decoction can alleviate the apoptosis,but Qileng Decoction is preceded to Buyanghuangwu Decoction. Concolusion:Qileng Decoction play the neuron protective effects by restraining the expression of the Caspase-3 protein and reducing the neuron apoptosis.
