摘要
从半夏中提取基因组DNA作为模板进行RAPD反应的优化试验,获得RAPD-PCR扩增反应的最佳条件:25μL体系中Mg2+1.5 mmol/L;dNTPs各0.2 mmol/L;模板DNA100 ng;引物0.25μmol/L;Taq酶活性值1.0 U。
Genomic DNAs Pinellia ternata was used as template to optimize the random amplified polymorphic DNA ( RAPD ) reaction system , and some essential factors which might take effects on the results of RAPD were investigated. The optimization of a 25ul RAPD-PCR system was determined as followed: 1.5 mmol/L Mg^2+ ,0.2 mmol/L dNTPs for each, 100ng template DNA, 0.25μmol/L primer and 1.0 U Taq DNA polymerase.
