烟草K326叶片愈伤组织诱导及继代的初步研究 被引量:6

摘要 以烟草K326叶片为外植体,采用MS基本培养基,研究了不同激素组合对愈伤组织的诱导效果,以及继代培养过程中不同浓度的NAA和6-BA对愈伤组织生长的影响。研究结果表明:2.4-D2.0 mg/L,6!BA0.5 mg/L组合诱导效果最好,7d后叶块边缘长出愈伤组织,诱导率为100%。不同浓度NAA和6-BA组合的愈伤组织生长速度、结构、质地和颜色有显著差异。以NAA0.1 mg/L+6-BA3.0mg/L、NAA2.0mg/L+6-BA0.5 mg/L和NAA1.5 mg/L+6-BA0.5mg/L3种培养基组合的愈伤组织生长较快。其中前一组合愈伤组织为绿黄色,结构致密,质地硬;后两种组合愈伤组织为黄白色,结构疏松,质地脆。 The leaf of tobacco variety-K326 was used as the explants on the MS basal medium and the influence on callus induction by different hormones, as well as the influence on its subculture with the combination of NAA and 6-BA were studied. The results showed that 2.4-D 2.0 mg/L + 6-BA 0.5 mg/L was the most effective combination for callus induction. During the subculture, the different concentration of NAA + 6-BA combination had different effect on callus growth, structure, texture, color and luster, and the combinations of NAA O. 1 mg/L + 6-BA 3.0 mg/L, NAA 2.0 mg/L + 6-BA 0.5 mg/L and NAA 1.5 mg/L + 6-BA 0.5 mg/L were preferable. Callus on the medium with NAA 0. 1 mg/L + 6-BA 3.0 mg/ L were the green color with yellow, tightness and hard. Callus on NAA 2.0 mg/L + 6-BA 0.5 mg/L and NAA 1.5 mg/L + 6-BA 0.5 mg/L were the yellow color with white, loose and soft.
