逆灸对佐剂性关节炎大鼠保护效应的抗氧化机制研究 被引量:3

摘要 目的:探讨逆灸对完全弗氏佐剂关节炎(AA)大鼠的保护作用及抗氧化机制。方法:大鼠大椎穴逆灸(预先艾灸)。用AA大鼠模型,观察逆灸对AA大鼠早期和继发期原发关节组织病理学及下丘脑、血浆中超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量的影响。结果:与正常组比较,AA各组大鼠原发足踝关节滑膜炎症表现更为明显,早期重于继发期,继发期关节软骨呈现部分损坏。同时,AA各组大鼠血浆和下丘脑SOD活性下降(P<0.05或P<0.01);MDA含量增加(P<0.01)。与同期AA各组比较,逆灸可使原发足踝关节的组织病理学得到明显的改善,并使血浆、下丘脑SOD活性明显回升(P<0.05或P<0.01),血浆MDA明显下降(P<0.05),但仅使继发期AA大鼠下丘脑MDA有所下降(P<0.01),对早期下丘脑MDA影响不大(P>0.05)。逆灸还可使正常大鼠血浆SOD活性显著提高(P<0.05)。结论:逆灸对AA大鼠早期和继发期原发关节炎具有阻抑减轻的保护作用,这可能与逆灸调整早期及继发期血液及下丘脑自由基代谢失衡有关。 To study on protective effect and antioxidative mechanism of preventive moxibustion in rats with adjuvant arthritis (AA). Methods: Preventive moxibustion was applied at Dazhui (GV 14) in AA rats, and the histopathologieal changes of the joints, the activity of superoxide dismutase (SOD) and malondialdehyde (MAD) content in hypothalamus and plasma were investigated in the rats at the early and secondary stages. Results:As compared with the normal group,primary synovitis of the ankle joint was more obvious in the various AA groups ,and at the secondary stage ,the articular cartilage showed a part of injury. SOD activities decreased and MDA content increased in plasma and hypothalamus of the various AA groups(P〈0.05 or P〈0.01). The preventive moxibustion could improve significantly histipathological changes of the primary ankle joints,increase the activities of SOD in plasma and hypothalamus(P〈0. 05 or P〈0. 01) and decrease MDA level in plasma (P〈0.05) and the hypothalamus(P 〈0. 01) of AA rats at the secondary stage (P〈0.0l ), but did not significantly change MDA level in the hypothalamus (P 〉 0. 05 ) at the early stage, as compared with the various AA groups at the same stage. The preventive moxibustion could significantly increase plasma SOD activity in the normal rat (P〈0.05). Conclusion: Preventive moxibustion has protective action of inhibiting and relieving primary arthritis at the early and secondary stages in AA rats,and this action is possibly related with preventive moxibustion regulating metabolism of free radicals in blood and the hypothalamus at the early and secondary stages.
