摘要
根据黄牛β-珠蛋白基因的序列设计引物,扩增了中国牦牛的β-珠蛋白基因,并对其进行了克隆测序和氨基酸序列比较分析。结果显示,其与黄牛β-珠蛋白基因的同源性很高,达到97%以上;检测到中国牦牛β-珠蛋白基因的2个等位基因7β3A sn和1β35A sn,其中67号牦牛个体中检测到2个等位基因7β3A sn和1β35A sn,64号牦牛个体中检测到等位基因1β35A sn,1078号牦牛个体中检测到等位基因7β3A sn;等位基因1β35A sn是中国牦牛特有的一个等位基因,推测该等位基因编码的第135位氨基酸天冬酰胺,可能会降低牦牛血红蛋白的氧亲和力。
Yak β globin is amplified by primers designed from cattle β globin gene. The cloning and sequencing results indicate that they have very high homology with cattle β-globin gene with a value of above 97%. Two alleles,β^72Asn and β^135Asn,were detected at β-globin locus in Chinese yak. The sample 67 has these two alleles,while individual 64 has allelc β^135Asn and individual 1078 has allele β^73Asn. The allele β^135Asn is the characteristic of Chinese yak after residue blasting. It is suggested that the β^135 Asn of this allele could reduce the oxygen affinity of yak hemoglobin.
