新疆红胫戟纹蝗痘病毒包涵体蛋白基因序列分析

摘要 通过对红胫戟纹蝗痘病毒Dociostarus kraussiEPV(DkEPV)包涵体蛋白基因克隆、测序与同源性分析,结果显示,DkEPV包涵体蛋白基因包含2 922 bps的开放阅读框架,编码109.4 kDa蛋白质。与鳞翅目及鞘翅目昆虫痘病毒包涵体蛋白氨基酸的同源性小于20%,而与其他直翅目昆虫痘病毒包涵体蛋白氨基酸的同源性均高于80%。DkEPV包涵体蛋白包含19个半胱氨酸位点,主要分布在C-末端。此包涵体蛋白基因启动子区域基因,富含A+T,并且具有典型的痘病毒晚期启动子信号TAAATG,直翅目昆虫痘病毒之间此序列保守。同时在此痘病毒包涵体基因的下游克隆了另一个不完整的基因序列,与血黑蝗痘病毒的MSV072基因同源,与包涵体蛋白基因比邻但方向相反。 The spheroidin gene of Dociostarus kraussi EPV(DkEPV)was cloned, sequenced and analyzed. In the DkEPV spheroidin gene, a ORF of 2 922 bps was harbored that it was capable of encoding polypeptide of 109. 4 kDa. Computer analysis indicated that the spheroidin shared less than 20 % aa identity with lepidopteran EPVs and eoleopteran EPVs spheroidins, but more than 80 % aa identity with orthopteran CiEPV, GsEPV, OaEPV, MsEPV and AaEPV spheroidins. The spheroidin contained 19 cysteine residues that were particularly abundant at the C - temaini. The promoter regions of the spheroidin gene were highly conserved (99%) among the orthopteran EPVs and also contained the typical very A + T rich and TAAATG signal mediating transcription of poxvirus late genes. An incomplete ORF downstream of the pheroidin gene was also sequenced. The ORF was in the opposite direction to the spheroidin gene and was homologous to the MSV072 putative protein of MsEPV.
