摘要 目的生物节律广泛存在于人类生活和工作中,为研究正常人节律基因表达特点,定量检测正常年轻人外周血中节律基因Per1(humanPeriod1,hPer1)的表达。方法实验对象为7名大学生(H1-H7),年龄24 ̄30岁,男性3名,女性4名。其中H7于实验前1天晚上值夜班。分别在09:00,15:00,21:00,03:00,09:00点抽取肘静脉血6mL,抽提全血总RNA。应用竞争性RT-PCR方法定量检测实验对象外周血中每人每个时间点hPer1基因的表达。结果人类外周血中hPer1基因的表达有显著节律性。表达高峰在09:00而低峰在21:00,两者之间的差异达2.29±0.9倍(P<0.01),第2天09:00hPer1基因表达水平又回到最高峰。结论hPer1基因的表达在外周血中存在节律性。 [Objective] To investigate circadian rhythm of human Perl gene expression in peripheral mononuclear system(PMS)in healthy young adults and changes of Perl gene expression after work shift. [Methods] 7 healthy young adults (H1-H7), aged from 24-30, 3 male and 4 female, were recruited. H7 had work shift just before experiment. Competitive RT-PCR methods were used to quantify gene expression. [Results] Human Perl gene expression was signifieandy higher at 9:00 clock and reached the lower level around 21:00. The difference between the higher peak and lower peak of hPerl expression was significant (P 〈0.01). Perl gene expression at 9:00 clock in the first day was approximately equal to that of the second day(P 〉0.05). [Conclusions] There is a significant circadian rhythm in the expression of hPerl in human PMS. These results would contribute to the study of circadian rhythm in patients and explore the relationship between circadian rhythm and diseases.
