α黑素细胞刺激素在TREM-1介导的树突状细胞成熟中的调节作用

摘要 研究α黑素细胞刺激素(-αMSH)对髓样细胞表达的激发受体1(TREM-1)介导树突状细胞(DC)成熟的调节作用。用TREM-1激动性抗体处理单核细胞,诱导其分化成MoDC,在此过程中加入-αMSH。观察MoDC的细胞形态,以FACS进行表型测定,用混合淋巴细胞反应检测MoDC的抗原提呈功能,并观察-αMSH对MoDC表达TREM-1的影响。结果显示,-αMSH和TREM-1激动性抗体作用MoDC后,多数细胞突起少且钝,α-MSH对TREM-1激动性抗体处理的MoDC高表达MHC II、CD86及CD1a有抑制作用,并降低MoDC刺激同种异体T细胞增殖的能力。-αMSH可下调LPS诱导MoDC表达TREM-1的水平。上述结果表明,α-MSH能抑制TREM-1介导的MoDC成熟。 The regulatory action of α-melanocyte stimulating hormone(α-MSH) on the dendritic cell maturation mediated by triggering receptor expressed on myeloid cells 1 (TREM-1) was investigated. Monocyte-derived dendritic cell (MoDC) was induced by agonist anti-TREM-1 mAb, which was then cultured with α-MSH. The morphological change was observed, and the phenotype was detected using FACS. The antigen presenting function of MoDC was tested with mixed lymphocyte reaction. The effect of α-MSH on the TREM-1 expression of MoDC was evaluated. The results showed that in comparison with anti- TREM-1-qreated MoDC, α-MSH+anti-TREM-1 treated MoDC had less and blunt dendrites and down regulation of CD86, CDla and HLA DR. α-MSH inhibited the MoDC stimulated proliferation of alloreactive T cells. LPS-induced TREM-1 expression on MoDC was also inhibited by α-MSH. These results demonstrate that α-MSH is able to block the MoDC maturation mediated by TREM 1.
