骨形成蛋白4对大鼠牙乳头细胞生物学行为的影响

摘要 目的观察骨形成蛋白4(BMP4)对体外培养的大鼠牙乳头细胞增殖及分化的影响,为探讨BMP4在牙胚发生发育中的调控作用奠定基础。方法体外培养胚胎13d的大鼠牙乳头细胞,利用MTT法和测定细胞内碱性磷酸酶含量检测BMP4对大鼠牙乳头细胞增殖及分化的影响。结果BMP4能抑制牙乳头细胞的增殖,并能促进细胞分泌碱性磷酸酶(P<0.01)。结论BMP4具有抑制大鼠牙乳头细胞增殖,并促进牙乳头细胞成熟分化的调控作用。 Objective To observe the effects of bone morphagenetic protein 4(BMP4) on proliferation and differentiation of dental papilla cell of rat and explore the role of BMP4 in regulation of tooth germs development.Methods Dental papilla ceils extracted from rat embryo aged 13 days were cultured in vitro.The proliferation and differentiation effects of BMP4 on the cells were examined by MTT method and level detection of alkaline phosphatase of the cells.Result BMP4 could significantly inhibit dental papilla cells proliferation and stimulate ALPase activity (P〈0.01).Conclusion BMP4 plays an important regulation role in inhibiting proliferation and promoting differentiation of dental papilla ceils.
