摘要 目的研究大肠癌VEGF-C和VEGFR-3 mRNA的表达与大肠癌淋巴管生成的关系。方法采用RT-PCR检测32例新鲜大肠癌及相应正常组织中VEGF-C和VEGFR-3 mRNA表达,免疫组化染色淋巴管内皮特异标记物podoplanin计数淋巴管密度,以血管标记物CD34染色做对比。结果大肠癌VEGF-C及其受体VEGFR-3 mRNA水平显著高于正常组织,且二者表达具有显著正相关性。大肠癌VEGF-C mRNA高表达组癌周淋巴管密度显著高于低表达组。伴有淋巴管受累或淋巴结转移组VEGF-C mRNA显著高于无淋巴管受累或淋巴结转移组。结论VEGF-C/VEGFR-3信号转导途径可能对大肠癌癌周淋巴管生成具有重要作用,并可能进而促进癌组织淋巴结转移。 Objective To study the relationship of expression of VEGFC and VEGFR-3 mRNA with lymphangiogenesis in colorectal cancer. Methods The mRNA expression of VEGF-C and its receptor VEGFR-3 in 32 cases with CRC were detected by RT PCR. The microlymphatic density (MLD) and microvessel density (MVD) were evaluated by immunohistoehemistry with antihuman podoplanin and CDa4 antibody respectively, Results VEGF-C and VEGFR-3 mRNA level of tumor tissues was significantly higher than that in normal mucosa. Moreover, the mRNA expression of VEGFC was significantly coincident and correlated with that of VEGFR-3. VEGF-C mRNA expression was significanlly correlated with MLD at the edge of CRC and with lymphatic involvement and lymph nodes metastasis in CRCs. Conclusion VEGF-C/-D-VEGFR-3 signal pathway might play an important role in peritumoral lymphangiogenesis, and promote lymph node metastasis.
