大鼠腰椎终板软骨细胞的培养及其生物学性状的研究 被引量:5

摘要 目的通过对大鼠腰椎终板软骨细胞的培养,观察细胞的生物学性状,探讨终板软骨细胞的生物学行为及影响因素。方法取大鼠腰椎终板软骨细胞,在加10%灭活胎牛血清的F12-DMEM培养液中培养,建立体外终板软骨细胞培养模型。采取绘制细胞生长曲线、HE、甲苯胺蓝染色及免疫组织化学染色等方法,对细胞形态、活力、生长情况及软骨细胞的表型进行鉴定。结果终板软骨细胞可以在体外培养,但细胞活力随着培养时间的延长及传代逐渐降低;终板软骨细胞的生长情况及细胞表型类似于关节软骨,有Ⅱ型胶原和aggrecan的表达。结论体外成功培养了大鼠腰椎终板软骨细胞,为进一步研究终板软骨在椎间盘退变中的作用打下了基础。 Objective To find a method of lumbar endplate cartilage cells culture and to observe its biological charactenstics. Methods The cells were isolated from lumbar endplate cartilage, then cultured in F12- DMEM with 10ml/L fetal bovine serum. By HE and Toluidine blue staining and by immunocytoehemistry for Ⅱ type collagen and aggrecan, the biological characteristics and morphology of the cells were identified. Results The cell's phenotype and growing model was similar to articular cartilage cells. The expression of Ⅱ type collagen and aggrecan was positive. Conclusion In vitro, the rat's lumbar endplate cartilage cells could be cultured , it would be benefit to study the mechanism of intervetebral discs degeneration.
