聪脑汤对实验性阿尔茨海默病大鼠单胺类神经递质的影响 被引量:2

摘要 目的探讨聪脑汤对实验性阿尔茨海默病(AD)大鼠模型学习记忆能力的影响及作用机制。方法60只大鼠随机分为正常组、假手术组、模型组、聪脑汤大、中、小剂量组。采用D-半乳糖合并鹅膏覃氨酸损毁大脑基底核神经元的方法复制模型。各组动物以通道式水迷宫游出时间和错误次数为学习记忆评价指标,水迷宫试验结束后各组测定脑海马单胺类神经递质含量的变化。结果与正常组与假手术组相比,模型组动物水迷宫游出时间和错误次数显著升高,海马多巴胺(DA)、去甲肾上腺素(NE)、5-羟色胺(5-HT)含量水平明显下降。与模型组相比,聪脑汤大、中、小剂量组游出时间和错误次数均显著降低,同时能明显升高海马DA、NE、5-HT水平,其中以高剂量组作用更明显。结论聪脑汤能改善模型大鼠的学习记忆功能,其作用机制与提高AD模型大鼠海马DA、NE、5-HT含量有关。 Objective To observe the effect of Congnao decoction on activity of Dopaminergic Neuro - transmitter in Rat Dementia Model. Methods Sixty male SD rats were randomized into normal group, sham operation control group, model group, Congnao decoction treatment groupl (lower dosage), group 2 (medium dosage) and group 3 (high dosage). Basal forebrain- injured rat was used as model of Alzheimer' s disease. Water maze was applied in six groups. The activity of dopaminergic neuro- transmitter in brain was detected. Results Compared with normal group and sham operation control group, the swimming time and the error number were obviously prolonged and the level of dopaminergic neuro- transmitter in brain was decreased significantly in model group. The swimming time and error number were shortened, the levels of dopaminergic neuro- transmitter in brain were increased in three treatment groups compared with model group. Conclusion Congnao decoction could improve the ability of learning and memory, increase cerebral dopaminergic neuro- transmitter in brain of model rat, exerting a positive effect on treatment of cognitive dysfunction.
