摘要
目的:通过对TNF及铜绿假单胞菌(PAO1)刺激肺上皮细胞株(A549)表达胞浆蛋白NOD2的研究,进一步了解NOD2在机体天然免疫病原体识别过程中的作用。方法:培养呼吸道肺癌上皮细胞株A549,以TNF、铜绿假单胞菌PAO1刺激细胞,并以未刺激组做对照,通过半定量RT-PCR技术观察胞浆蛋白NOD2的表达变化。结果:与未刺激组比较,TNF、铜绿假单胞菌PAO1都能够上调A549细胞NOD2蛋白的表达。结论:TNFα与铜绿假单胞菌PAO1刺激能增强A549细胞胞浆蛋白NOD2的表达,提示细胞内受体NOD2在呼吸道天然免疫病原体识别过程中可能起重要作用。
Objective: The aim of this study was to describe the regulation of NOD2 expression in airway epithelial cells. Methods: Expression of NOD2 mRNA was determined by reverse-transcription polymerase chain reaction (RT-PCR). Results:TNF and Pseduomonas aeruginosa PAO1 both induce an up-regulation of NOD2 expression in epithelial cell line A549. Conclusion:Tile treatment of TNFα and Pseduomonas aeruginosa up-regulates the expression of the NOD2 gene in respiratory epithelial ceils ,which suggest that NOD2 may serve as an innate immune receptor recognizing respiratory pathogens.
