摘要
从多个农药厂含氯污泥及附近土样中筛选出数株对2-氯丙酸具有较高脱氯能力的菌株,经过脱氯曲线和生长曲线的考察,获得了活性和稳定性均好的菌株。通过超声波破碎细胞方法的正交实验,确定了超声时间3 s,间歇时间5 s,超声次数70次,破碎功率600 W。对从该菌中分离提取的粗酶液进行了酶学性质考察,得到脱卤酶反应的适宜条件是pH为7.5,30 ℃。脱卤酶在pH为7.0-7.5时最稳定,在35 ℃时酶活最高,但热稳定性较差。
Some strains had been screened from the sludge and the soil of some pesticide factories. All of them had high dehalogenation activities to 2-CPA, in which strain 9 #: showed high activity and good stability. The extraction process of crude enzyme from strain 9 #: by ultrasoniation was observed. According to orthogonal test, optimum ultrasonic parameters were ultrasonic time 3 second, intermittent time 5 second, 70 ultrasonic times and ultrasonic power 600 W. It was investigated that the enzymatic characteristics of the crude enzyme extracted from strain 9 # showed optimum reaction pH and temperature were 7.5 and 30 ℃ respectively. The enzyme showed good stability in pH= 7. 0-7. 5 and high enzyme activity at 35 ℃, but the enzyme thermostability appeared worse at 35 ℃.
