摘要
缝隙连接通道是连接相邻心肌细胞的特殊通道,其主要功能是介导心肌细胞间化学信息的交换和心肌细胞间的电耦联,其表达和功能的正常是心脏整体正常电活动的重要保证。心脏缝隙连接通道由不同的连接蛋白组成,心室的缝隙连接通道主要由连接蛋白43组成。心脏缝隙连接通道的功能受胞内pH值、胞内Ca2+浓度、ATP浓度、连接蛋白的磷酸化状态、跨通道电压和一些神经体液因子等的调控。近年的研究发现,心肌细胞内存在一些能够与连接蛋白43存在相互作用的蛋白质,并且发现这些蛋白质能够通过这种相互作用改变连接蛋白43的结构状态或磷酸化状态,从而进一步发挥调控缝隙连接通道的功能。现仅就近年来发现的与Cx43存在相互作用的蛋白质及其调控作用综述如下。
Gap junctions(GJs) are special channels connecting the adjacent cardieeytes . The gap junction's fundamental functions arc chemical and electrical communication between the my,cordial cells. The normal expression and function of gap junction are essential to the normal electric activity of the whole heart. The GJs in the heart are composed of different connexins(Cxs), the connexin43 (Cx43) is the predominant protein consisting in the gap junction localized in ventricle. The function of the cardiac GJ is regulated by different factors including intracellular PH, concentration of intracellular Ca^2 + and ATP, the phosphorylation status of the Cxs, the voltage difference between two end of the channel, some neurohumoral factors and etc. Recent studies have shown that some proteins in cardiocytes can interact with Cx43 and regulate the function of the gap junction channel by changing the configuration and/or phnsphorylation status of Cx43. In this review,we summarized the proteins which can interact with Cx43 and regulate the function of gap junction channel.
