摘要 目的探讨肾小球细胞凋亡对肾结构与肾功能的影响研究。方法建立阿霉素肾硬化大鼠模型,光镜和电镜观察肾小球结构变化,并检测其有核细胞记数。用原位检测肾小球细胞凋亡指数。结果疾病组各时间点调亡指数明显升高(P<0.01),于12周达高峰。疾病组肾小球中有核细胞数与肾小球细胞凋亡指数之间存在明显负相关。肾小球内凋亡细胞数与肾功能恶化程度呈密切正相关, 凋亡指数与 BUN 和 Scr 的相关系数分别为0.8543和0.9006。结论随病情进展,实验性肾硬化大鼠中肾小球细胞发生过度调亡,导致细胞数目减少,加重肾小球硬化进展。 Objective To study the effect of glomerular cell apoptosis on renal stracture and renal finction in rat with glomerulosclerosis.Methods Chronic glomerulosclerosis was induced by intravenous injection of amycin in the rats undergoing uninephectomy. Blood urea nltrogen(BUN) ,serum creatinine(Scr)and 24 hour urine protein were determined. The glomerular nuclear cell counts were performed on HE staining slices. The Apoptosis of the glomerular cells was assessed by in situ dUIP blot nick - end labehng method(TUNEL).Results The number of apoptotic glomemlar cells gradually increased after the intravenous injeceion of amycin and peaked 12 weeks after the injection. The apoptotic cells were 1.92±0.52,3.50±0.68,10.03± 1. 67 respectively the 4th,8th and 12th weeks after the injection in the expermental group.Them was significantly negative correlations between the glomerular cell apoptosis index and the number of nuclear cells (r =-0.7456, P 〈0.01).There was significantly positive corrrelations between apoptosis index and BUN or Scr(r = 0.8543 and 0.9036 respectively, P 〈 0.01),Conclusions The excessive apoptosis of glomerular cells occurs in the rats with glomerulosclerosis and deteriorates the glomerulosclerosis .
