摘要
目的应用电针刺激SD大鼠肝俞、足三里、丰隆、太冲穴,观察其对高脂饮食所致的NASH大鼠血清TNFα含量及IR水平的影响。方法40只SD大鼠随机分为正常对照组、模型组、电针组和西药组,每组10只大鼠,在16周后隔夜空腹以2%戊巴比妥钠(45mg/kg)麻醉,经下腔静脉采血,分离血清。采用OlympusAu560全自动生化分析仪检测血清ALT、AST、TNFα、FBG等;采用放射免疫分析法测定空腹血清胰岛素,计算胰岛素抵抗指数。结果ALT、AST、TNFα模型组比正常组显著增高(t分别为4.4990、3.2929、-7.7330,P<0.01);模型组比电针组显著增高(t分别为3.9218、3.0284、-8.0031,P<0.01);电针组较之西药组有下降趋势,但二者比较无明显差异(t=1.1650、2.4512、0.3448,P>0.05)。空腹血糖正常组与其他各组比较,差异无统计学意义(P>0.05);胰岛素抵抗指数水平模型组比正常组显著增高(t=-2.9310,P<0.01);模型组比电针组增高(t=-2.2161,P<0.05);电针组较之西药组有下降趋势,但二者比较无明显差异(t=0.4574,P>0.05)。相关分析表明,在模型组、电针组和西药组IR与TNFα水平均呈正相关(r分别为0.9024、0.3464、0.5512)。结论非酒精性脂肪性肝炎大鼠血清肿瘤坏死因子α(TNFα)过度表达,并且与IR的程度呈正相关,二者共同参与NASH的发生;电针可能通过抑制TNFα的高度表达而起到改善IR达到治疗NASH的目的。
Objective:To observe the effects of electrical acupuncture on serum TNF-α and IR in the livers of rats with Non-alcoholic Steatohepatitis, Acupoints Ganshu (BL 18), Zusanli (ST 36), Fenglong (ST 40) and Taichong (LR 3) were needled with electrical acupuncture. Methods: 40 SD rats were randomly divided into 4 groups of ten each: normal group, model group, electrical acupuncture group and the western medicine group. The rats were hoeused with 2% pentobarbital sodium (45mg/kg weight) at the end of the 16^th week from the beginning of the experiment. The whole ,blood was gathered through inferior vena eava and serum was collected. Serum ALT, AST, TNFα and FBG were tested with Olympus Au560 automatic biochemistry analyzer and tasting serum insulin was tested with the method of radio immunoassay. Then insulin resistance index was calculated, Results: Serum ALT, AST, TNFα were markedly increased in the model group in comparison to the normal group ( t=4, 4990, 3. 2929 and -7. 7330 respectively, P〈0. 01 ); So were in the model group in comparison to the electrical acupuncture group (t= 3. 9218, 3. 0284 and -8. 0031 respectively, P〈0. 01) ; There was a downtrend concerning serum ALT, AST, TNFα in the electrical acupuncture group comparing with those in the western medicine group but there was no statistical difference between them (t=1. 1650, 2. 4512 and 0. 3448 respectively, P〉0. 05). There was no significant difference between the four groups with regard to fasting blood glucose (P〉0. 05) ; IR level was markedly higher in the model group than that in the normal group (t=-2. 9310, P〈0.01); it was higher in the model group than that in the electrical acupuncture group (t=-2. 2161, P〈0. 05) ; There was a downtrend in the electrical acupuncture group in comparison to the western medicine group, but there was no significant difference (t=0. 4574, P〉0. 05) . Correlation analysis showed IR was in positive correlation to Serum T N F a in the model group, electrical acupuncture group and the western medicine group (r = 0. 9024, 0. 3464, 0. 5512 respectively). Conclusion: the overexpression of serum TNFα of the rats with NASH is in positive correlation to the levels of IR. TNFα and IR both play important roles in the development of NASH;Electrical acupuncture might treat NASH effectively through its inhibitory effects on the overexpression of serum TNFα which results in the improvement of IR.
