摘要
Iq7611是从中草药马蔺子的种皮中分离出来的一种醌类化合物。本实验以碱洗脱微孔滤膜法证明了Iq7611(20、40μg/ml)能导致小鼠白血病L1210细胞DNA单链断裂,这可能是Iq7611的抗癌作用机理之一。实验还证实,博莱霉素也能导致DNA单链断裂,但细胞经37℃温育后,其DNA断裂仍可修复。而Iq7611(10μg/ml)与BLM合用则可抑制BLM所致的DNlA单链断裂后的重接修复。
Iq7611 was isolated from the coating of seeds of Iris pallassi Fisch, Var Chinensis. It has a quinone structure. By the modified alkaline elution, we demonstrated that Iq7611, at the concentration of 20, 40μg/ml, could induce DNA single strand break in L1210 leukemia cells. This effect my be one of the antitumor mechanisms of Iq7611. Our results also showed that bleomycin ( BLM ) , at 7.5, 15 and 3(μg/ml, could induce DNA single strand breaks, but the breakage would be repaired after incubation of the cells in 37℃. However, Iq7611 (10μg/ml) combined with BLM at above concentration could inhibit the rejoining of DNA single strand breaks induced by BLM.
