摘要
本文从P^(21)高表达质粒PJCL.E_(30),分离出P^(21)蛋白,免疫后,取睥细胞与小鼠骨髓瘤细胞SP 2/O融合,制备了高效价P^(21)单克隆抗体。以此单抗为探针,应用免疫组化技术,对50例鼻咽部组织ras—P^(21)的表达进行了探测。结果表明,在鼻咽癌组织中具有明显阳性表达(阳性率为85%),而鼻咽部炎症仅1例有表达,正常胎儿鼻咽组织未观察到阳性表达。说明在鼻咽癌变中,当ras基因被激活,其表达增强,P^(21)蛋白量增加。本文首次报道了P^(21)在鼻咽癌中具有阳性表达,这不仅对研究ras基因在鼻咽癌变中的作用,并对临床应用,均具有重要的理论意义及实用价值。
The ras P21 protein was transformed and purified from high expressionrecombinant plasmid PJCL-E30, and was immunized with BALB/c mice. The spleen cells of immunized mice were fused with BALB/c myeloma cell line Sp2/0 Ag 14. High efficient P21 monoclonal antibodies was chosen from five hybridomas. Using the MAb-P21 as a probe, the expression of gene product was studied by immunohistochemical technique (ABC method).The expression of MAb-P21 was detected in 50 cases of naspharyngeal tissues, among them 40 cases were nasopharyngeal carcinoma (NPC), 7 cases were nasopharyngitis, 3 cases were normal fetal nasopharynx. Positive expression of the ras P21 protein was observed obviously in NPC and the positive rate was 85% (34/40). The immunoreactivity was heterogeneity in cytoplasmic ras P21 expression. The ras P21 was located in the inner side of the cell membrane and the positive expression was shown to be deep yellow stained in cell section. There was no detectable ras P21 evpression in the normal fetal nasopharyngeal tissues, and for nasopharyngitis there was only one case (1/7) expressed positively.These results implicated that activated ras gene is one of the transforming genes of NPC, and that in the carcinogenic process of human nasopharyngeal carcinoma the gene expressisnowas enhanced.
