摘要
目的:研究野生型K-ras2基因对人结肠癌细胞株Caco-2的生长影响作用。方法:将野生型K-ras2基因转染结肠癌细胞株Caco-2并筛选稳定表达的克隆;用四甲基偶氮唑(MTT)法观察细胞的生长情况,用流式细胞仪观察细胞周期及凋亡的情况。结果:野生型K-ras2基因可使结肠癌细胞株Caco-2的生长明显受到抑制,使细胞受阻于G0/G1期。结论:野生型K-ras2基因能有效抑制结肠癌细胞Caco-2的生长。
Objective:To observe the growth inhibitary effect of wild-type K-ras2 gene on colon adenocarcinoma cell line Caco-2. Methods:The colon adenocarcinoma cell line Caco-2 was transfected with the wild-type K-ras2 gene, and stable expression clones were selected. The growth of the colonadenocarcinoma cell was detected by monotetrazolium(MTr) , and the cell cycle analysis was carried out by flow cytometry (FCM). Results:The expression of wild-type K-ras2 gene observably suppressed the growth of the colon adenocarcinoma cell line Caco-2 and arrested cell cycle in the G0/G1 phase. Conclusion:The wild-type K-ras2 gene effectively suppressed the growth of colon adenocarcinoma cell line Caco-2.
