慢性坐骨神经损伤大鼠海马GFAP和pERK表达的变化

摘要 目的:探讨慢性坐骨神经挤压损伤(CC I)大鼠海马组织中胶质原纤维酸性蛋白(GFAP)和磷酸化有丝分裂素激活蛋白激酶(pERK)表达的改变。方法:按Bennett法制作CC I模型,采用免疫组化法测定CC I大鼠海马组织中GFAP和pERK的表达。结果:CC I大鼠双侧海马齿状回胶质细胞GFAP和pERK表达均有增加,以手术对侧增加更为显著,与假手术组比较差异有显著性(P<0.01)。结论:中枢神经系统胶质细胞ERK/MAP激酶通路激活,可能参与慢性神经痛的发病过程。 Objective:To determine the expression of glia fibrillary acidic protein(GFAP) and phosphorylated extraeellular signal-regulated kinase(pERK) in the hippocampus of chronic constriction injury(CCI) rats. Methods:The rats were placed four loose ligatures around the right sciatic nerve and the changes of GFAP and pERK protein expression in the hippocampus was detected by immuno-histochemieal techniques. Results: Immunoreactivity (IR) of GFAP, pERK protein was significantly increased in the contralateral hippocampus in CCI group(P 〈 0.01 ). Conclusion: CCI may activate ERK/MAP (Mitogen activated protein) kinase pathways in astrocytes in the hippocampus, and ERK/MAPK pathways may play an important role in the genesis of neuropathic pain.
