甘草酸对慢性马兜铃酸肾病大鼠尿蛋白与尿酶排泄及肾功能的影响 被引量:13

摘要 目的观察甘草酸对慢性AAN肾损害的干预作用。方法将雄性Wistar大鼠随机分为正常组、模型组、甘草酸组、泼尼松组。正常组灌胃饮用水;实验组按马兜铃酸20mg.kg-1.d-1灌胃关木通浸膏,建立大鼠慢性AAN动物模型,并于2h后按分组灌胃给药:甘草酸25mg.kg-1.d-1,泼尼松3.15mg.kg-1.d-1,模型组灌胃饮用水。各组每周测量体重,于第4、8、12周代谢笼法留取24 h尿液,腹主动脉采血并分离血清,测定相关肾功能指标。结果随着喂饲时间延长,正常组大鼠体重稳定性增长,而马兜铃酸造模各组体重增长缓慢,尿白蛋白、NAG酶排泄量、肌酐/体重水平均明显升高,其中甘草酸组、泼尼松组尿白蛋白和NAG酶排泄量、血肌酐/体重水平均较模型组明显降低。结论甘草酸对马兜铃酸肾损害有一定的保护作用,能降低尿白蛋白和尿ANG酶排泄,改善肾功能。 Objective To study the effects of Diammonium giycyrrihizinate on the urine albunfin, NAG enzyme excretion, and renal function in the rats of chronic aristolochic acid nephropathy. Methods Male wistar rats were divided into control group, model group, DG group, Prednisone group at random. The control group was given potable water; the test groups were firstly given the Extracta of Caulis Aristolochiae Manshuriensis(CAM) (aristolochic acid ,AA20 mg·kg^-1·d^-1) intragastically respectively. Two hours later, the DG group was given DG 25 mg·kg^-1·d^-1, the Prednisone group Prednisone 3.15 mg·kg^-1·d^-1, model group equal amount of potable water. The rats' body weights were measured weekly. At the end of 4th , 8th and 12th week, take their 24h-urine, blood serum samples from abdominal artery, for detection of indexes of renal functions, Results Along with feeding times, the control group body weight increased continuously, but the test groups' body weight increased slowly. The urine albumin, NAG enzyme excretion, the level of serum creatinin increased markedly after giving AA in the test groups, but declined in therapeutic groups. These mean that the AA impaired the renal function, but the DG and Prednisone had some protective effects. Conclusion DG and Prednisone could reduce the urine albumin, NAG enzyme excretion and the level of serum creatinin, with some protective effects on the chronic aristolochic acid nephropathy.
