翼状胬肉中端粒酶催化基团与凋亡相关基因蛋白的表达 被引量:4

摘要 目的探讨端粒酶逆转录酶催化亚基(telom erase reverse transcriptase,hTRT)和抑凋亡基因bcl-2、凋亡基因bax产物在翼状胬肉发病机制中的作用。方法应用链霉抗生物素蛋白-过氧化物酶免疫组织化学方法,检测20例翼状胬肉、9例正常结膜组织中hTRT、bcl-2、bax基因产物的表达。结果hTRT和bcl-2在翼状胬肉中的阳性表达率显著高于正常结膜(P<0.01),主要表达于翼状胬肉的上皮基底层细胞,正常结膜无表达;bax在翼状胬肉与正常结膜中的表达无显著差异。hTRT与bcl-2表达呈正相关。结论翼状胬肉的发生、发展与hTRT和bcl-2、bax的异常表达有关,可能是细胞异常增殖和凋亡失衡的共同结果。 Objective To study the roles of telomerase reverse transcriptase (hTRT) ,bcl-2 genes and bax gene products in the pathogenesis of pterygium. Methods The expressions of bcl-2,bax and hTRT were detected in 20 pterygium and 9 normal eonjunctival spectimens by immunohistochemical streptavidin-peroxidases(SP). Results The expressions of hTRT and bcl-2 in pterygium were significantly higher than those of normal conjunctiva( P 〈 0.01 ). hTRT and bcl-2 staining noted predominantly in the basal epiththelial layer of cells, but no evidence was found in normal conjunctival specimens. There was no difference in the expressions of bax between pterygium and normal conjunctiva. The expression of hTRT was correlated with that of bcl-2. Conclusion The abnormal expressions of hTRT and bcl-2 may be positive factors in the pathogenesis of pterygium. The results suggest that pterygium development may result from a co-operation of abnormal preliferation and unbanlanced apoptosis.
