摘要
首先测定8种蔬菜细胞壁中的羟脯氨酸含量,研究结果表明:豆皮、豆荚、花粉、胡萝卜根、油菜叶柄、芹菜叶柄、青菜和洋葱细胞壁中羟脯氨酸的含量分别为6.02μg/mg、3.30μg/mg、1.74μg/mg、1.53μg/mg、0.64μg/mg、0.52μg/mg、0.42μg/mg和0.31μg/mg。然后通过CM-纤维素柱和Sephadex G-75柱色谱纯化豆渣细胞壁中的伸展蛋白。用CM-纤维素柱色谱纯化豆渣细胞壁蛋白时,得到3个组分,即S1、S2和S3;对S2组分进一步采用Sephadex G-75柱色谱纯化,得到S2a、S2b和S2c,并对S2c组分进行单糖组分分析,S2c中的单糖以阿拉伯糖和半乳糖为主,它们的摩尔之比为21∶。
The amount of hydroxyproine in the cell wall from eight kinds of vegetables were determined in the present paper. The result showed that the amount of hydroxyproine in the cell wall from skin of soja, legume, pollen, carrot root, cole, celery, green grocery and onion was 6.02μg/mg, 3.30μg/mg, 1.74μg/mg, 1.53μg/mg, 0.64μg/mg, 0.52μg/mg, 0.42μg/mg and 0.31μg/mg, respectively. The extensin from the cell wall of soybean dregs were purified by CM-cellulose and Sephadex G-75 chromatography. Three components were attained from the cell wall proteins of soybean dregs by CM-cellulose chromatography, they were S1, S2 and S3. S2 was separated into S2a, S2b and S2b by Sephadex G-75 chromatography. The mono-saccharide composing of S2c was analyzed by GC spectra, the mole ratio of arabinose and galactose is 2:1.
