中风康对局灶性脑缺血大鼠脑组织bFGFmRNA表达的影响

摘要 目的探讨中风康对局灶性脑缺血大鼠脑组织碱性成纤维细胞生长因子(bFGF)mRNA表达的影响。方法采用线栓法复制局灶性脑缺血大鼠模型,采用RT-PCR技术于脑缺血6,12,24,48h动态观察局灶性脑缺血大鼠脑组织bFGFmRNA表达的变化,并在缺血24h以步长脑心通为对照,观察中风康对bFGFmRNA表达的影响。结果与假手术组比较,局灶性脑缺血6h,模型组脑组织bFGFmRNA表达量开始增加,至24h达到高峰(P<0.01);与模型组比较,脑缺血24h各治疗组脑组织bFGFmRNA表达量均有不同程度升高(P<0.05或P<0.01),以高剂量组升高最为显著(P<0.01)。结论中风康可在转录水平提高脑梗死大鼠脑组织bFGFmRNA表达,有效减轻局灶性脑梗死引起的缺血性损伤。 Objective To observe the effect of Zhongfengkang on the expression of basic fibroblast growth factor(bFGF) mRNA in brain of focal cerebral ischemia rats. Methods The rat model of focal cerebral ischemia was established by inserting nylon thread, the expression changes of bFGFmRNA in brain of focal cerebral ischemia rats were observed by RT-PCR in 6, 12, 24 and 48 h after cerebral ischemia. The effect between Zhongfengkang and Buchangnaoxintong on the expression of bFGFmRNA in brain of focal cerebral ischemia rats was compared 24 h after cerebral ischemia. Results The expression of bFGFmRNA in brain of model group increased 6 h after cerebral ischemia, and reached top at 24 h(P 〈0. 01 )compared to that of sham group;the expression of bFGFmRNA in brain of each treatment group were increased ( P 〈 0. 05 or P 〈 0. 01 ) compared to that of model group, and that of high-dose group were increased most remarkedly ( P 〈 0. O1 ). Conclusions Through increasing the expression of bFGFmRNA in brain of focal cerebral ischemia rats on transcriptive level, Zhongfengkang could protect focal cerebral ischemia rats effectively.
