摘要
目的探讨不同浓度三氧化二砷(As2O3)对食管癌细胞系EC9706的生长抑制作用及其机理。方法不同浓度三氧化二砷作用于细胞EC9706,采用MTT法检测细胞生长,倒置显微镜观察细胞形态,并通过DNA梯状电泳和流式细胞仪检测凋亡。结果三氧化二砷剂量依赖性抑制细胞生长;观察到典型的细胞凋亡形态学特征性改变;琼脂糖电泳呈现凋亡DNA梯状条带;流式细胞仪分析,细胞株EC9706存在明显的G0/G1期阻滞。结论As2O3具有抑制食管癌细胞株EC9706生长作用,其机理是诱导细胞凋亡和细胞周期阻滞。
Objective To observe the effect of a wide range of concentration of Arsenic Trioxide on esophageal cancer cell EC9706. Methods The cell growth activity was detected by MTT assay and the morphologic changes was studied after treatment with different concentration. The apoptosis was detected by flow cytometry assay and DNA Gel electrophoresis. Result The cell growth was depressed by Arsenic Trioxide depending on concentration obviously. The presented apoptosis features were observed; DNA Gel electrophoresis showed DNA ladder; Flow cytometry analysis showed an arrestment at G0/G1 phase. Conclusion Arsenic Trioxide can inhibit the growth of esophageal cancer cell EC9706 obviously through inducing cell apoptosis and arrestment at G0/G1 phase.
