骨髓基质细胞源神经干细胞对局灶性脑缺血大鼠行为及神经细胞凋亡的动态影响 被引量:5

摘要 目的探讨骨髓基质细胞(BMSCs)源神经干细胞对局灶性脑缺血大鼠行为及神经细胞凋亡的影响。方法建立大鼠大脑中动脉缺血再灌注模型。32只健康Sprague-Dawley(SD)大鼠分为假手术组、缺血对照组、缺血骨髓基质细胞移植组和缺血骨髓基质细胞源神经干细胞移植组。分别于移植后1d、7d、14d、28d对大鼠进行神经功能缺损评分和空间学习能力检测,并在移植后14d和28d分别应用TUNEL法检测神经细胞凋亡。结果移植后7d各组神经功能均有不同程度的恢复,BMSCs源神经干细胞移植组大鼠的神经功能评分[(6.3±2.32)分]显著优于BMSCs移植组[(7.1±1.88)分]。移植14d后可提高大鼠空间学习能力,且各时间点BMSCs源神经干细胞移植组的空间学习能力改善优于BMSCs组。随移植后时间的延长,BMSCs源神经干细胞移植组和BMSCs移植组的神经功能和空间学习能力更趋改善。BM-SCs源神经干细胞移植组缺血边缘区凋亡细胞数[(4.8±1.12)个]明显少于BMSCs移植[(6.3±2.73)个]。结论骨髓基质细胞源神经干细胞移植可改善大鼠神经行为、减少神经细胞凋亡,显著优于BMSCs移植。 Objective To study the effects of bone marrow stromal cells derived neural stem cells on be- havior and celluar apoptosis after focal cerebral ischemia and reperfusion. Methods The model of middle cerebral artery occlusion and reperfusion were established. 32 Sprague-Dawley rats were divided into sham-operated group, ischemia control group, BMSCs transplanted group and BMSCs derived neural stem cells transplanted group . The neurogenic behavior of the rats were evaluated in 1 ,7,14,28 days after transplantation . The rats were killed separatedly in 14,28 days after transplantation. The brain sections were used for TUNEL staining. Results The neurological function of rats were recovery in 7 clays later after transplantation . The recovery of neurological function in BMSCs derived neural stem cells transplantation group (6.3±2.32 ) was evidently than BMSCs transplantation group(7.10±1.88). The abilities of spatial learning and memory of rats in 14 days later after transplantation got improved, what's more , had more satisfactory outcome in BMSCs derived neural stem cells transplanted group than in BMSCs transplanted group. Abilities of spatial learning and memory of rats in both BMSCs derived neural stem cells and BMSCs transplanted group got continuously improved along with the time elapsed. The number of apoptotic cells in the group of BMSCs derived neural stem cells transplanted group (4.80±1.12 )decreased evidently compared with that of the BMSCs transplantation group(6.30±2.73). Conclusion BMSCs derived neural stem cells may make the neurogenic behavior recovery, decrease the infarct volumes and the number of apoptotic cells in cerebral ischemia rats.
