摘要
探讨P44/42MAPK在X射线诱发会厌癌的表达及其作用。从X射线诱发的会厌癌中得到肿瘤组织及相应的癌旁正常组织,分离总蛋白及mRNA,应用蛋白印迹法分析两组P44/42MAPK的表达及磷酸化水平,应用逆转录-聚合酶链反应(Reversetranscription-polymerasechainreaction,RT?PCR)半定量方法得到两组P42MAPK的mRNA变化。结果表明,会厌癌P44/42MAPK的蛋白表达及磷酸化水平均显著高于癌旁正常组织,P42MAPK的mRNA在会厌癌中的表达也明显升高。结果显示,P44/42MAPK在X射线诱发会厌癌有异常表达且磷酸化水平显著增高,此信号转导途径可能在X射线诱发会厌癌发挥一定的作用。
The work is to investigate the role of the P 44 / 42 mitogen-activated protein kinases in proliferation disorder of epiglottis carcinoma induced by X-ray irradiation. ARer obtaining the tissues from the radiation-induced epiglottis carcinoma, we observed the P 44/42 MAPK protein expression level and the phosphoryation level in epiglottis carcinoma by western blotting, and the mRNA level by semi-quantitative RT-PCR. The results showed that The P 44 / 42 MAPK expression of protein and mRNA was significantly higher in lepiglottis carcinoma than in normal tissue nearby the tumor. And the level of P44/42MAPK phosphoryation in epiglottis carcinoma was higher than in that of the normal tissue. The results suggest that P44/42 MAPK may play a role in proliferation disorder of epiglottis carcinoma I induced by the radiation.
