摘要 研究电离辐射对体外培养肾小管上皮细胞1α羟化酶表达的影响。采用实时荧光定量(Real?timeQuantitativePolymeraseChainReaction,Real?timeQ?PCR)技术与透射电镜等方法,观察3Gy、5Gy和10Gy137Csγ射线单次照射(剂量率为0.85Gy/min)体外培养肾小管上皮细胞后1α羟化酶mRNA表达水平及细胞线粒体形态的改变,并与不受照(0Gy)组比较。结果表明,低剂量(3Gy)γ射线照射24h后即出现肾小管上皮细胞1α羟化酶mRNA表达水平的明显降低(p<0.05),且随受照剂量的增加下降幅度增大;电镜观察显示受照后肾小管上皮细胞线粒体呈现肿胀、基质变薄、电子密度减低、嵴断裂、数量减少与排列紊乱等变化。 The work was to study in vitro the effects of ionizing radiations on the expression of 1α-hydroxylase in renal tubular epithelial cells, which were exposed to 3Gy, SGy and 10Gy by ^137Cs T-rays. By using Real-time Q-PCR technology and transmission electron microscopy (TEM), changes in the 1α-hydroxylase mRNA level and the mitochondria ultrastructure after the irradiation were compared with the control (0 Gy) group. An obvious decrease in the 1α-hydroxylase mRNA expression was found after the 3 Gy irradiation, and the decreasing rate increased with the irradiation dose. The TEM results also showed that with the irradiated cells the mitochondria swelled obviously, the electron density decreased, and the cristae arrangement breaked and disordered.
