摘要
利用细胞培养技术,研究胰岛素浓度为10、100、1000和10000mU/L时对各组别兔主动脉平滑肌细胞之细胞动力学、形态学变化。结果表明:①胰岛素浓度为10mU/L时,细胞动力学和形态学均无明显变化。②胰岛素浓度≥100mU/L时,细胞明显增殖,且DNA增殖率和胰岛素浓度呈正相关(r=0.53,P<0.01)。③在胰岛素作用下(≥100mU/L),平滑肌细胞呈高电子密度(合成型)及低电子密度(收缩型)二种形态学改变;部份平滑肌细胞内出现脂滴,后者有利于泡沫细胞形成。本文结果提示:降低或避免糖尿病患者的高胰岛素血症,可延缓或减轻动脉粥样硬化的发生与发展。
The effect of insulin concentrations with 10,100,1 000 and 10 000 mU/L on the cytodynamics and cytomorphology of cultured rabbit arterial smooth muscle cells(SMC) was carried out by isotope technique, light microscopy and transmission electron microscopy. The results were as follows:(1)No effect of insulin concentration 10 mU/L on SMC in both cytodynamics and cytomorphology was found.(2) Insulin concentrations with≥100 mU/L stimulated multiplication of SMC, and positively related to the multiplication rate of DNA, a parameter of cytodynamics ( r=0.53,P< 0.01 ) .(3) In insulin concentrations≥100 mU/L groups two types of cytomorphological changes, high(synthetic type) and low(contractile type) electronic density of SMC, were observed. Furthermore, lipidic substance in some SMC was found under such concentration of insulin, which was favorable of formation of foam cells. Our results suggest that it may delay the development of atherosclemosis to keep lower levels of serum insulin or avoid hyperinsulimemia in diabetic patients.
