摘要
用细胞培养方法观察了不同浓度(10-9~10-5mol/L)的全反式维甲酸(ATRA)对正常造血祖细胞集落形成的影响。结果表明,随着浓度的增加,ATRA对红系祖细胞集落形成单位(CFU-E、BFU-E)的抑制作用增加,但在10-8~10-6mol/L内对粒单系祖细胞集落形成单位(CFU-GM)的形成有促进作用,10-6mol/L的ATRA对巨核系祖细胞集落形成单位(CFU-Meg)生长也有促进作用,提示在急性早幼粒细胞白血病(APL)用ATRA治疗过程中,ATRA参与了其正常造血功能的恢复。
Using cell culture method, we investigated the effects of concentration of all trans retinoic acid (ATRA) (10 -9  ̄10 -5 mol/L) on the formation of hematopoietic progenitor cell in normal human bone marrow. Constant suppression on CFU E and BFU E colony formation and coloring was observed in a dose related fashion from 10 -9  ̄10 -5 mol/L ( n =3). ATRA enhanced the total number of granulocytic/macrophage and megakaryocytic colony between concentration of 10 -8 and 10 -6 mol/L and 10 -6 mol/L, respectively. These results suggest ATRA may play a role in the recovery of hematopoiesis during treatment of acute promyelocytic leukemia with ATRA.
