抗脲激酶单克隆抗体的研制和初步鉴定

Establishment and Characterization of Monoclonal Antibody vs Urokinase

用上海生物化学制药厂赠送的脲激酶（Ｈ－ＵＫ），免疫ＢＡＬＢ／Ｃ小鼠，取脾细胞与骨髓瘤细胞ＮＳ－１融合，用高纯度Ｈ－ＵＫ包被筛选阳性克隆，获得抗Ｈ－ＵＫ单克隆抗体ＮＵＫ－１和ＮＵＫ－２两细胞株。酶联免疫吸附试验（ＥＬＩＳＡ）检测效价为１０４～１０５，经亚类测定分属ＩｇＧ１和ＩｇＧ２。为研究脲激酶的结构和导向治疗血栓提供了可能。

Anti H UK monoclonal antibodies, NUK 1 and NUK 2 cells, were obtained by means′ of screening with high purity H UK coated positive clones. Using ELISA, the products revesled a titre of 10 4￣10 6, belonging to subgroups IgG1 and IgG2, respectively. H UK, a gift from Shanghai Biochemical Pharmaceutical Plant, was used to immune BALB/C mice. Splenocytes and myelomal NS 1 cells were fused in the preparation. This preparation provides a possibility for the clucidation of the structure of urokinase and drug directed treatment of thrombosis.
