刺五加对大鼠脑缺血损伤修复作用的研究 被引量:13

摘要 采用双侧颈总动脉结扎制作SD大鼠暂时性全脑缺血模型,手术后尾静脉分别注射药物刺五加注射液或生理盐水进行观察和神经行为学评分,制片作常规HE染色和Nestin免疫组化染色检测缺血损伤后的修复情况.神经行为学评分显示刺五加组神经功能恢复明显好于生理盐水组.脑缺血再灌注后3d时海马、室管膜和室管膜下区神经干细胞有少量Nestin阳性表达;脑缺血再灌注后5d和7d时海马、室管膜、室管膜下区和缺血区神经干细胞阳性Nestin阳性细胞数明显增加,尤其在缺血区,刺五加组Nestin阳性表达细胞数显著多于生理盐水对照组.结果表明刺五加有促进脑缺血脑组织神经干细胞的增殖,对大鼠脑缺血损伤有一定修复作用. To investigate the effects of acanthopanax senticosus on the proliferation of the neural stem cells after cerebral ischemia reperfusion in SD rats, the temporary cerebral ischemia model was established by common carotid arteries occlusion. The acanthopanax senticosus or saline were injected into rats through trail vein after operation. The behavior of the operated rats were observed and the neurologic scores 2 hours after ischemic reperfusion were assessed respectively. The slices were stained with hematoxylin and eosin and the immunohistochemical staining was performed to observe the changes of nestin protein level in ischemic region. The results of neurologic scores revealed that the effect of acanthopanax senticosus was better than that of saline group. The nestin protein was found to be expressed in a few neuronal stem cells 3d after ischemia in the hippocampine zone , ependymal zone and subependymal zone, but the number of neuronal stem cells increased obviously 5-7d after ischemia. Especially in the ischemic region, the number of neuronal stem cells was signifi- cantly more than that in the saline group. Conclusion Acanthopanax senticosus can promote the proliferation of neuronal stem cells in the tissue of cerebral ischemia and improve the damage after cerebral ischemia in SD rats.
