一株高羊茅内生真菌的分离、鉴定以及对油菜菌核病原菌抗性的初步研究 被引量:6

Identification of an Antimicriobial Endophytic Fungus Associatied with Tall Fescue and It's Antagonism on Sclerotinia sclerotiorum

摘要 从高羊茅(Fescue arundinacea)植株中分离纯化得到6株内生真菌,通过抑菌谱的研究发现J24对油菜菌核病原菌Sclerotinia sclerotiorum有抑制作用.体外对峙实验观察两菌落之间有明显的拮抗带.圆盘滤膜法研究发现其非挥发性代谢产物可以抑制核盘菌菌丝的生长,抑制率达到69%;平板上形成菌核的重量减少率为49.7%,菌核萌发率为69.2%.培养基分割法研究发现可挥发性产物可以抑制菌核的萌发,培养7d菌核萌发率为0.J24在生物防治方面存在巨大的潜力.对J24进行体外培养及电镜观察,分生孢子长卵形,(4.1~7.2)×(1.2~1.5)μm.初步鉴定为Acremonium alternatum,与高羊茅优势内生真菌Neotyphodium coenophialum不同。从而发现了一个新的共生体系. 6 endophytic fungi were isolated and puridied from Festuca arundinacea . Dual cultural assay on pota- to dextrose agar revealed that J24 was antagonistic to Sclerotinia sclerotiorum. It's non-volatile metabolites suppressed mycelial growth of S. sclerotiorum and decreased biomass of sclerotia produced on the plates. And its volatile metabolites can also suppressed germination of sclerotial . According to observation of morphological characters,the isolate had relatively high similarity with Acremonium alternatum, which different from Neo- typhodium coenophialum.
