摘要 目的:探讨生物可降解纤维蛋白胶体作为恶性骨肿瘤化疗药物载体的可能性。方法:对骨肉瘤细胞株进行培养,采用MTT法测定化疗药物及不同浓度纤维蛋白对骨肉瘤细胞的抑制率,相对敏感药物梯度浓度的肿瘤抑制率,敏感药物与纤维蛋白联合应用对肿瘤的抑制率。结果:①单纯低、中浓度纤维蛋白对骨肉瘤细胞在体外有轻度抑制作用,高浓度则有促进瘤细胞生长作用。②低、中浓度纤维蛋白与盐酸阿霉素联合应用的抑瘤率比单纯盐酸阿霉素的抑瘤率低,但无统计学差异(P>0.05),高浓度纤维蛋白则有促进瘤细胞生长作用。纤维蛋白与顺氯伯(CDDP)联合应用的抑瘤率随纤维蛋白浓度升高而增高,各组间差异显著(P<0.05)。纤维蛋白与甲氨蝶呤(MTX)联合应用在高浓度组抑瘤率明显增高(P<0.05)。结论:纤维蛋白胶可作为CDDP、MTX而不是阿霉素(ADM)的良好缓释载体,并可增强前两者的药效。纤维蛋白胶与敏感药物制成的缓释系统用于局部治疗可在降低术后复发率上起到积极的作用。 Objective: To determine the use of fibrin glue as a drug carrier in treatment of osteosareoma. Methods:The specimens of osteosarcoma were cultured. Then, in vitro chemosensitives of fibrin glue and fibrin glue with adriamycin (ADM), cisplachitin (CDDP) or methotrexate (MTX) to osteosareoma were tested using MTY method, Results: 1, only low and middle concentrations of fibrin had modest inhibitory effect on cells of osteosareoma. However, the high concentration had stimulation effect. 2. Although the inhibitory rates of low and middle concentration fibrin with ADM were lower than that of ADM in equal concentration (maximum concentration in plasma) , this was not significant (P 〉 0.05 ). The high concentration showed the stimulation of growth of tumor cells. The inhibitory rates of three different concentrations of fibrin glue with CDDP were higher than that of CDDP in equal concentration (maximum concentration in plasma) , demonstrating a positive correlation with fibrin concentration (P 〈 0.05). Furthermore, the high concentration of fibrin glue with MTX showed the highest rate of inhibitory (P 〈 0.05 ). Conclusion:Fibrin glue can be used as a good sustained release carrier of MTX or CDDP, except ADM. Sensitive chemical drug-loaded fibrin glue delivery system may be used for local treatment of malignant bone tumor to reduce the recurrent rate after operation.
