摘要
目的研究离体培养的人中性粒细胞在被LPS激活的情况下,槲皮素对中性粒细胞产生IL-6的影响。方法运用ELISA法检测槲皮素对LPS诱导的中性粒细胞分泌IL-6的影响;运用流式细胞术检测槲皮素对LPS活化的中性粒细胞内IL-6的蛋白水平的影响;运用逆转录PCR(RT-PCR)技术检测槲皮素对LPS活化的中性粒细胞表达IL-6 mRNA的影响。结果在无刺激的情况下中性粒细胞不产生IL-6,LPS(1μg/mL)以时间依赖性的方式诱导中性粒细胞表达IL-6 mRNA,合成并分泌IL-6,并在16 h达到高峰。然而,预先用槲皮素(40μmol/L)处理中性粒细胞30 min后,LPS诱导的中性粒细胞表达IL-6 mRNA,合成并分泌IL-6则受到了抑制。结论IL-6是炎症早期重要的促炎症因子。因此,槲皮素抑制LPS诱导中性粒细胞产生IL-6,提示槲皮素可能通过对炎症因子负性调控而发挥其抗炎作用。
Objective To invistigate the effect of quercetin on lipopolysaccharide (LPS)-induced IL-6 production by neutrophils. neihods The level IL-6 in the culture supematants of LPS-stimulated neutrophils was assessed by ELISA before and after quercetin treatment; the level of IL-6 protein in LPS-stimulated neutrophils was detected by flow cytometry (FACS) ; the IL-6 mRNA expression of LPS-stimulated neutrophils was assayed by RT-PCR. Results Resting neutrophils did not express IL-6, and LPS ( 1 μg/mL) could induce neutrophils to express IL-6 in a time-dependent manner. After 30 min pre-treatment with quercetin (40 μmol/L), the LPS-induced IL-6 production by neutrophils were inhibited. Conclusion Quercetin might exerts anti-inflammatory effects through negatively modulating pro-inflammatory factors, such as IL-6. The inhibitory effects of quercetin on IL-6 production by neutrophils may provide a theoretic basis on future therapy of inflammation.
