摘要
目的研究SP2/0细胞膜抗原负载树突状细胞(DC)诱导的特异性细胞毒性T淋巴细胞(CTL)对肿瘤细胞的杀伤活性。方法密度梯度离心法分离SP2/0细胞膜,负载经rmGM-CSF和rmIL-4诱导扩增的小鼠骨髓来源的DC,活化T淋巴细胞获得肿瘤特异性CTL,MTT法检测对SP2/0细胞的杀伤效果。结果骨髓单个核细胞在rmGM-CSF和rmIL-4作用下获得了高表达CD80、CD86和MHC-Ⅱ分子的DC,致敏的CTL对SP2/0骨髓瘤细胞具有高杀伤率,显著高于对L5178Y淋巴瘤细胞的杀伤活性(P<0.01)。结论SP2/0细胞膜抗原负载DC能诱导明显的特异性抗肿瘤免疫应答。
Objective To investigate the cytotoxic efficiency of tumor specific cytotoxic T lymphocyte (CTL) induced by SP2/0 cellular membrane antigen-loaded syngeneic dendritic cells (DC). Methods SP2/0 cellular membrane was obtained by density gradient centrifugation. Bone marrow-derived DC Balb/c of inbred mouse hne was generated in vitro in the presence of granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-4 (IL-4). The tumor-specific CTL was induced by membrane antigen-loaded DC. The cytotoxicity of CTL was assayed by MTT. Mouse lymphoma cell line L5178Y was used as control in this experiment. Results In the presence of GM-CSF and IL-4, the bone marrow precursors of Balb/c mouse generated DCs that highly expressed major-histocompatibility-complex class Ⅱ (MHC- Ⅱ ) and costimulating molecules CD80 and CD86. CTLs induced with SP2/0 cellular membrane antigen-loaded DCs had strong tumor-specific cytotoxicity to SP2/0 cells, especially at the high effector/target ratio. These CTLs had very low cytotoxic effects to L5178Y lymphoma cells compared with SP2/0 cells ( P 〈 0.01). Conclusion SP2/0 cellular membrane antigen-loaded DC can elicit potent specific antitumor immune response in vitro.
