摘要
目的:探讨半胱氨酸蛋白酶-8(Caspase-8)在膀胱癌发生、发展中的作用及其意义。方法:应用免疫组化Elivision法检测Caspase-8在正常膀胱组织、腺性膀胱炎、膀胱癌旁组织、膀胱移行细胞癌及膀胱腺癌中的表达;用图像分析系统测量Caspase-8的平均光密度(OD)值。结果:Caspase-8在上述组织中的平均OD值依次为(0.248±0.044)、(0.150±0.051)、(0.234±0.053)、(0.108±0.053)、(0.105±0.062)。膀胱癌中Caspase-8的表达明显低于正常膀胱组(P<0.01),Caspase-8在膀胱移行细胞癌中的表达随病理分级上升而减弱(P<0.01),且与分级存在明显的相关性(r=-0.674;P<0.01);但与病理分期无关。结论:Caspase-8在膀胱癌中表达的下调,揭示膀胱癌的演进过程与Caspase-8有密切关系。
Objective:To study the role of Caspase-8 in the development of carcinoma of bladder, as well as its clinical significance. Methods: The expression of Caspase-8 were evaluated immunohistochemically in the normal bladder samples,cystitis glandularis, paraneoplastic samples and carcinoma of bladder by measureing the average optical density with image analysis system. Results: The average optical density of Caspase-8 in normal bladder samples,cystitis glandularis, paraneoplastic samples, transitional cell carcinoma of bladder and adenocarcinoma of bladder was respectively (0. 248±0. 044),(0. 150±0. 051),(0,234±0. 053),(0. 108±0. 053),(0. 105±0. 062). Successively the expression of Caspase 8 in carcinoma of bladder was significantly lower than that in normal bladder samples ( P〈0.01). The intensity of Caspase-8 expression was decreased with the increasing of grades of tumor ( P 〈0. 01), and obviously correlated with the grades ( γ=-0. 674; P〈0.01), but incorrelated with stages of tumor. Conclusions:The down regulation of Caspase-8 expression in bladder neoplasm reveals the close relationship between Caspase-8 and the occurrence and progression of the cancer.
