摘要
利用鸡胚性腺生殖细胞-体细胞体外无血清共培养模型研究雄激素对生殖细胞增殖的影响。培养的鸡胚卵巢细胞用睾酮(T,10-8、10-7、10-6mol/L)和/或芳香化酶抑制剂letrozole(Let,10-9、10-8、10-7mol/L)处理,48h后测定生殖细胞增殖的变化。结果显示,睾酮能够促进卵巢生殖细胞的增殖,且这种促增殖作用可被Let部分阻断。由此推断,睾酮的这种促进鸡胚卵巢生殖细胞增殖的作用,部分是通过转变为雌激素才得以发挥的。
Effect of androgen on embryonic chicken ovarian germ cell proliferation was evaluated by a germ-somatic cell coculture system. Cultured cells were challenged with testosterone(10^-8, 10^-7 10^-6 mol/L) and an aromatase inhibitor letrozole (10^-9 10^-8,10^-7 mol/L) for 48 h and then the germ cell proliferation was determined. Results showed that testosterone stimulated proliferation of embryonic chicken germ cells and this effect could be partially prevented by letrozole. These results indicated that testosterone stimulate proliferation of cultured ovarian germ cells should be partially via conversion to estrogen in embryonic chickens.
