摘要
目的观察大小鼠癌组织内的毛细淋巴管的形态特征,并探讨肿瘤组织内是否有淋巴管新生,为研究癌的淋巴道转移机制提供形态学依据。方法构建大鼠结肠癌和小鼠移植瘤模型,取材后进行5′-Nase-ALPase双重染色,透射电镜观察肿瘤内毛细淋巴管超微结构。结果肿瘤的周边区的毛细淋巴管多于组织正常区,肿瘤中心无毛细淋巴管,毛细淋巴管壁上可见破坏和开放连接,同时观察到淋巴管内皮细胞亚细胞结构的改变,在移植瘤内可见从一些较大的毛细淋巴管上伸出小的管腔。结论肿瘤细胞可通过破坏的淋巴管壁或内皮细胞间的开放连接侵入淋巴管腔,细胞器的改变是内皮细胞损伤的结构基础,在移植瘤内发现的毛细淋巴管可能为新生淋巴管,它们通过质膜小泡引导物质的转运。
Objective:To study the morphological features of lymphatic capillaries in tumors of rats and mice and discuss whether there is lymphangiogenesis during tumor growth. Methods:Colonic carcinoma and transplanted tumors were induced in rats and mice, respectively. The tumor tissue was removed, sliced and double-stained with 5'-Nase and ALPase. The superstructure of lymphatic capillaries in tumors was observed by TEM. Results:There were more lymphatic capillaries in the peripheral part of tumors than that in the normal tissues. There were no lymphatic capillaries in the central part of tumors. The wall of lymphatic capillaries wa.s destroyed and open junction of lymphatic endothelial cells was found. The organdies of lymphatic endothelial cells had some changes. Some'small lymphatic capillaries came up from the bigger lymphatic capillaries in transplanted tumors. Conclusions:Cancer cells may invade into the lymphatic capillaries through destroyed wall or the open junction of lymphatic endothelial cells. The changes o{ organdies are the basis of the injury of lymphatic endothelial cells. The lymphatic capillaries found in transplanted tumors may be de novo lymphatic capillaries which may help to lead substance transport by plasmalemmal vesicles.
