摘要
成熟虫卵是血吸虫病的主要致病因素和传播的唯一因子,抗卵免疫对于预防血吸虫病和阻断传播流行均具有重要意义。应用SMART cDNA文库构建技术,构建日本血吸虫虫卵全长cDNA文库。文库容量为7.55×106,滴度为7.2×1010,重组比率为97%。应用三个已知全长基因(SjPP,SjTEGT and SjeIF 5)引物在文库中成功钓取到相应的全长基因。另外还由文库直接扩增到一个日本血吸虫新基因烯醇酶基因。高质量的虫卵全长cDNA文库的构建,为抗病疫苗尤其是抗卵抗原的筛选和研究提供了重要资源。
Mature egg is the primary agent of pathogenesis in hosts and is responsible for dissemination of the parasite. Therefore, to develop anti-egg immunity is significant for preventing schistosomiasis and blocking transmission. Egg full length cDNA library of Schistosoma japonicum was constructed by using SMART cDNA library construction kit, The capacity of this egg library was 7.55 × 10^6, in which more than 97% clones are recombinant. Three known full length gene: SjPP, SjTEGT and SjeIF 5 were successfully amplified from egg cDNA library. Furthermore, a new full length gene, Enolase, was also successully fished. These datas show that egg full length cDNA library is of resonably good quality, and can be serve as the valuable material for screening the protective antigen genes, particularly anti-egg antigen genes.
