摘要
目的了解整合素在瘢痕挛缩过程中的作用。方法取10例增生性瘢痕标本进行成纤维细胞原代培养,通过荧光定量PCR法(FQ-PCR)测定经整合素和黏着斑激酶(FAK)抗体阻断后培养的瘢痕成纤维细胞中-αSMA mRNA表达量的变化。结果经整合素和FAK抗体阻断后培养的成纤维细胞-αSMA的基因拷贝数均较阴性对照组明显下降(P<0.05)。结论整合素及FAK通过促进-αSMA的合成引起瘢痕的挛缩,在瘢痕挛缩过程中具有重要的作用。
Objective To stud) the mechanism of integrin in hypertrophic scar. Methods fibroblasts from 10 samples of human hypertrophic scars was cultured, FQ-PCR assay was applied to detect mRNA expression of α-SMA in hypertrophic scar fibroblasts after integrin and FAK antibody blocking. Results mRNA of α-SMA in fibroblasts expressed obviously lower after integrin and FAK antibody blocking than that of control groups ( P 〈 0.05) . Conclusion Through accelerating the synthesis of α-SMA, integrin and FAK play an important role in contracture of hypertrophic scar.
