中孕期彩超辐照对胎鼠肾脏P53蛋白表达的影响 被引量:1

摘要 目的:研究彩色多普勒超声辐照中孕大鼠,对胎鼠肾脏P53蛋白表达的影响。方法:彩超辐照孕14dSD大鼠子宫体表投影区,按照射时间不同分为对照组、照射10min组、20min组、30min组。仪器采用Acuson公司Sequoia-512型彩色电脑声像仪,照射条件:4V1探头,二维频率H3.0MHz,彩色频率3.0MHz,热指数(TI)=1.8,机械指数(MI)=1.6。各组于既定照射结束后24h取胎鼠肾脏标本,HE染色光镜下观察肾脏组织结构,免疫组化方法检测P53蛋白的表达。结果采用统计软件SPSS11.0行多组均数比较的方差分析。结果:光镜观察各组胎鼠肾脏组织均未见明显损伤。免疫组化法检测,各组胎鼠肾小管上皮细胞内可见均匀散在的胞核棕黄色淡染的P53蛋白阳性表达。照射10min、20min组P53蛋白表达与对照组无显著差异(P>0.05);照射30min组P53蛋白表达增强,与对照组有显著差异(P<0.05)。结论:彩超辐照中孕期SD大鼠子宫投影区30min内,不会引起胎鼠肾脏明显的组织学损伤;照射30min,胎鼠肾脏P53蛋白表达增加。 Objective: To disclose the changes of protein P53 expression in fetal rats' kidneys when they were insonificated by diagnostic color Doppler ultrasound(CDUS) in the second-trimester. Methods: Pregnant 14 days SD rats were insonificated by diagnostic CDUS(Sequoia-512, transducer 4V1, H3.0MHz, TI=l.8, MI=I.6), and they were divided randomly according to insonification time(10min, 20min, 30min). Control group was sham-insonificated. The fetal rats' kidneys were resected 24 hours later. HE stain was done for microscopic observing, and protein P53 expressions in fetal rats' kidneys were examined by immunohistochemistry method. Image analysis was done with HPIAS-1000 pathology analyses system. Results: There were no pathologic changes under microscope. There were no significant changes in the 10min group and 20min group when they were compared with the control group(P〉0.05), but expression of protein P53 was significantly higher in the 30min group than that in the control group(P〈0.05). Conclusions: Insonifieation by diagnostic CDFI in the second-trimester within 30rain will pot cause histologie changes in fetal rats' kidneys under microscope, but insonifieation beyond 30rain can increase expression of protein P53 in fetal rats' kidneys.
